ACE-031 is a soluble fusion protein research compound consisting of the extracellular domain of activin receptor type IIB (ActRIIB) fused to the Fc portion of human IgG1. It functions as a decoy receptor that selectively binds to myostatin (GDF-8) and other TGF-β superfamily ligands, such as activins, thereby inhibiting their interaction with native ActRIIB receptors on muscle cells. In preclinical models, this blockade prevents myostatin-mediated negative regulation of muscle growth, resulting in increased skeletal muscle mass, fiber hypertrophy, and enhanced muscle strength independent of fiber type. ACE-031 is utilized in research to investigate mechanisms of muscle hypertrophy, therapeutic approaches to muscle-wasting conditions (e.g., muscular dystrophies, sarcopenia), and the broader roles of TGF-β signaling in muscle homeostasis and metabolism.
